Asymetrische Synthese und Untersuchung von Azepino[1,2-b]isochinolinen als Vorstufen zu Quinocarcin und Benzo[g]chinolino[2,3-a]chinolizidinen als Berberinanaloga

Diese Arbeit beschäftigt sich mit der Synthese von Azepino[1,2-b]isochinolinen als wichtige Schlüsselverbindungen für eine Totalsynthese des Tetrahydrosiochinolin-Alkaloides (-)-Quinocarcin sowie der Synthese und Untersuchung von Benzo[g]chinolino[2,3-a]chinolizidinen als Analoga des Isochinolin-Alkaloids Berberin. Im Hinblick auf die Totalsynthese von Quinocarcin konnten unterschiedlich substituierte Vorläufer synthetisiert werden und durch eine Lewis-Säure-katalysierte Carbonyl-En Cyclisierung stereoselektiv zu Azepino[1,2-b]isochinolinen umgesetzt werden. Auf diese Weise konnten in 1-Methoxymethyl- und 8-Hydroxy- substituierte Azepino[1,2-b]isochinoline synthetisiert werden. Benzo[g]chinolino[2,3-a]isochinoline besitzen ein um einen Piperidinring erweitertes Berberinmolekülgerüst und sollten auf eine dem Berberin analoge pharmakologische Wirkung getestet werden. Es wurden unterschiedlich konfigurierte und substituierte Chinolizidine synthetisiert, wobei Strukturelemente des Berberins eingeführt wurden. So konnte ein im Ring B aromatisiertes Chinolizidin synthetisiert und zum entsprechenden Chinolizidinyliumiodid oxidiert werden. Die Aktivität und Art der Wechselwirkung der Chinolizdine gegebenüber DNA wurde durch NMR Experimente von Substrat-Oligonukleotid Komplexen untersucht. Hierbei konnte die grundlegende Fähigkeit der Chinolizdine als Minor-Groove Binder an Oligonukleotide bewiesen werden.

This works deals with the synthesis of azepino[1,2-b]isoquinolines as key structural features for the total synthesis of the tetraydroisoquinoline alkaloid (-)-quinocarcine and secondly with the synthesis and evaluation of benzo[g]quinolino[2,3-a]quinolizidines as analoges of the isoquinoline alkaloid berberin. In view of the total synthesis of quinocarcine, differently substituted precursors were synthesized and furthermore cyclized to azepino[1,2-b]isoquinolines by a stereoselective lewis-acid-catalyzed carbonyle-ene cyclization. By this method the synthesis of 1-methoxymethyl and 8-hydroxy substituted azepino[1,2-b]isoquinolines was achieved. Benzo[g]quinolino[2,3-a]quinolizidines have the same molecular sceleton as berberin, expanded by a piperidine ring. Thus it was aimed to evaluate their pharmacological activity compared to berberin. Therefore quinolizidines with different configuration and substitution pattern were prepared implementing structural features of berberine. This way a quinolizidine with an aromatized ring B was prepared and oxidized to the corresponding quinolizidinyliumiodide. The activity and mode of action of the quinolizidines towards DNA was evaluated by nmr experiments of substrate-oligonucleotide complexes. Using this method the fundamental ability of the quinolizidines to act as minor groove binder could be proven.
